Also known as: Augmentin 625, Augmentin Duo, Augmentin 625 tablet
Augmentin 625 Duo is a prescription antibiotic tablet that contains amoxicillin 500 mg, a penicillin, and clavulanic acid 125 mg, which protects amoxicillin from enzymes made by resistant bacteria. It is used to treat certain bacterial infections, such as some ear, nose, throat, chest, urinary tract, skin and dental infections. It works only against bacteria and does not help colds, flu or other viral infections. Take it only when a doctor prescribes it, and complete the course.

What is Augmentin 625 Duo used for?
Augmentin 625 Duo is used for short courses to treat bacterial infections in adults and children over 12, when a doctor judges that the bacteria are likely to respond to it.
- Ear, nose and throat infections, such as sinusitis, middle ear infection and repeated tonsillitis
- Chest infections, such as flare-ups of chronic bronchitis or COPD and some types of pneumonia
- Urinary tract infections, such as bladder and kidney infections
- Skin and soft tissue infections, such as boils, abscesses, cellulitis and infected wounds
- Dental infections, such as a tooth abscess
- Some bone, joint and abdominal infections, and infections after childbirth or abortion, usually under hospital care
How does Augmentin 625 Duo work?
Augmentin 625 Duo combines two medicines. Amoxicillin, a penicillin antibiotic, stops bacteria from building their cell walls, so they burst and die. Some bacteria defend themselves by making beta-lactamase enzymes that destroy amoxicillin. Clavulanic acid blocks these enzymes, so amoxicillin can work against many bacteria that would otherwise resist it. The combination kills bacteria but has no effect on viruses.
What happens in your body after you take it
You swallow the tablet
Taken whole with water at the start of a meal, the film-coated tablet breaks down in the stomach and releases both medicines. Food helps clavulanic acid to be absorbed well and lowers the chance of an upset stomach.
Both medicines enter the blood
Amoxicillin and clavulanic acid are absorbed from the small intestine and reach their highest levels in the blood about 1 to 1.5 hours after the dose. Only a small part of each is bound to blood proteins, so most of it is free to spread into body tissues and fluids.
Clavulanic acid disarms the bacterial enzymes
At the site of infection, some bacteria release beta-lactamase enzymes that can break amoxicillin apart. Clavulanic acid binds to many of these enzymes and switches them off, so amoxicillin is not destroyed.
Amoxicillin breaks the bacterial wall
Amoxicillin attaches to penicillin-binding proteins, which bacteria use to build their cell walls. Without a strong wall, growing bacteria burst and die, and the immune system clears the infection over the following days.
Why the whole course matters
Each dose kills more bacteria, but some survive longer than others. Taking every dose for as long as the doctor prescribes helps clear the infection and lowers the chance that resistant bacteria are left behind.
Side effects in the gut
The medicine also kills some of the helpful bacteria that normally live in the gut and other parts of the body. This is why diarrhoea and thrush, a yeast infection of the mouth or vagina, are common side effects.
The kidneys clear it
Both medicines are removed quickly, mostly by the kidneys into the urine, and each has a half-life of about 1 hour. This is why doses are spaced through the day, and why people with kidney disease may need a different dose.
How to take Augmentin 625 Duo
Take Augmentin 625 Duo exactly as your doctor prescribes or as printed on the pack. The number of doses a day and the length of the course depend on the infection, your kidney function and your doctor's advice.
- It is usually taken twice a day, about 12 hours apart, and sometimes three times a day for more severe infections, as the doctor prescribes.
- Take it at the start of a meal, with a glass of water. Swallow the tablet whole; do not break or chew it.
- Complete the course your doctor prescribes, even if you feel better sooner. Stopping early can let the infection come back and adds to antibiotic resistance.
- If you miss a dose, take it when you remember, unless the next dose is nearly due. Do not take two doses together.
- The course is usually short. The label says treatment should not go beyond 14 days without a doctor's review.
- Augmentin 625 Duo tablets are not recommended for children aged 12 and under. Other forms with different strengths are made for children and are given only on a doctor's advice.
- Do not keep leftover tablets for later or share them with anyone else. An antibiotic should be taken only for an infection a doctor has diagnosed.
Side effects of Augmentin 625 Duo
Most people take Augmentin 625 Duo without serious problems, but side effects in the stomach and gut are common, and diarrhoea is the most frequent. You can report side effects to the Pharmacovigilance Programme of India helpline, 1800-180-3024.
Uncommon or mild
- Diarrhoea or loose stools
- Nausea or vomiting (less likely if it is taken at the start of a meal)
- Thrush: white patches in the mouth, or itching and discharge from the vagina
- Indigestion or stomach discomfort
- Mild skin rash or itching
- Headache or dizziness
Rare but serious: get medical help
- Signs of a severe allergic reaction: swelling of the face, lips, tongue or throat, wheezing, difficulty breathing or fainting. Get emergency help immediately
- A widespread rash with blisters, peeling skin, or sores in the mouth or eyes, or a rash with fever and swollen glands
- Severe or watery diarrhoea that does not stop, or diarrhoea with blood or mucus and stomach cramps, during the course or up to two months after it
- Yellow skin or eyes, dark urine, pale stools or pain in the upper right side of the tummy. Liver problems can appear up to several weeks after the course ends
- Repeated vomiting a few hours after each dose, with paleness or drowsiness, especially in children
- Chest pain together with signs of an allergic reaction
- Unusual bruising or bleeding, or fever with a rash, swollen glands and feeling very unwell, which can be signs of a rare blood or immune problem
- Passing very little urine, blood in the urine, or fits (seizures)
Who should be careful
Augmentin 625 Duo is not suitable for everyone. Tell your doctor before taking it if any of the following apply to you.
- Allergy to penicillin, amoxicillin, cephalosporins or any other antibiotic: it must not be taken by people who have had an allergic reaction to penicillins or cephalosporins
- Jaundice or liver problems in the past after taking amoxicillin with clavulanic acid: it must not be taken again
- Liver disease: the doctor may check your liver function during treatment
- Kidney disease: a lower dose or fewer doses a day may be needed
- Glandular fever (infectious mononucleosis): amoxicillin very often causes a rash in people with this viral infection, so it should be avoided
- Pregnancy: it is used in pregnancy only when a doctor judges it necessary. One study linked its use in women whose waters broke early, before labour, with a serious bowel problem in newborns
- Breastfeeding: small amounts pass into breast milk. Watch the baby for diarrhoea, thrush, rash or poor feeding, and tell your doctor
- Long courses: other germs such as yeasts can overgrow, and the doctor may check your blood, liver and kidney function
- Colds, flu and most coughs and sore throats are caused by viruses: this antibiotic does not help them
Medicines and things that interact with Augmentin 625 Duo
Augmentin 625 Duo can interact with some medicines. Tell your doctor or pharmacist about everything you take, including medicines bought without a prescription.
- Blood thinners such as warfarin or acenocoumarol: the blood can take longer to clot, so your INR may need extra checks
- Methotrexate: penicillins can slow its removal from the body and raise the risk of side effects
- Allopurinol (for gout): skin rashes become more likely
- Probenecid: raises amoxicillin levels; using them together is not recommended
- Mycophenolate mofetil (used after organ transplants): its level in the blood may fall, so the doctor may monitor you
- Birth control pills: the Indian label says antibiotics may make the combined pill less effective, and vomiting or severe diarrhoea can also stop the pill being absorbed. Ask your doctor whether you need extra contraception
- Urine sugar tests: some older urine glucose tests can give a false positive result
If someone takes too much
Taking more Augmentin 625 Duo than prescribed can cause stomach pain, vomiting and diarrhoea, with loss of body fluids. Rarely, very large doses can cause fits or crystals in the urine that harm the kidneys. If someone has taken too much, call 112 or go to the nearest hospital immediately and take the medicine pack with you.
Storage
Store at a temperature not above 25°C, in the original pack to protect the tablets from moisture, and out of the reach of children. Do not use after the expiry date.
Regulatory status in India
Prescription antibiotic sold in India. The combination of amoxicillin 500 mg and clavulanic acid 125 mg is not on the CDSCO list of prohibited medicines and was not among the 156 fixed-dose combinations banned in August 2024. Some other combinations containing amoxicillin are banned, for example amoxicillin with dicloxacillin and lactobacillus (2024) and amoxicillin with tinidazole (2016).
Other brands with the same medicine
Other brands with the same two ingredients at the same strength (amoxicillin 500 mg + clavulanic acid 125 mg) include Clavam 625 and Moxikind-CV 625. They work the same way. Augmentin 1g Duo contains more amoxicillin (875 mg), and children's syrups have different strengths. Check the label for the ingredients and strength, and do not switch without asking your doctor or pharmacist.
Other strengths and forms
Augmentin Duo is sold by GSK India as more than one tablet strength and as suspensions. Besides the 625 tablet (amoxicillin 500 mg with clavulanic acid 125 mg), there is a 1 g tablet (amoxicillin 875 mg with clavulanic acid 125 mg), and powders to make an oral suspension, such as Duo suspension and DDS, which have different strengths. Always check the label of the one you have; the strengths are not interchangeable.
Common questions
What is Augmentin 625 Duo used for?
It is an antibiotic used to treat certain bacterial infections, such as ear, nose and throat infections, chest infections, urinary tract infections, and skin and dental infections. A doctor prescribes it when bacteria are the likely cause. It does not help infections caused by viruses.
Does Augmentin 625 Duo work for a cold, flu or a sore throat?
Colds, flu and most sore throats and coughs are caused by viruses, and antibiotics do not work against viruses. Taking an antibiotic when it is not needed can cause side effects and helps bacteria become resistant. A doctor decides whether an infection is bacterial and needs an antibiotic.
What does 625 Duo mean?
The number 625 is the total of amoxicillin 500 mg and clavulanic acid 125 mg in each tablet. Duo refers to the twice-a-day dosing used for mild to moderate infections, about one tablet every 12 hours. For more severe infections a doctor may prescribe it three times a day.
Should I take Augmentin 625 Duo before or after food?
Take it at the start of a meal. Food helps the body absorb clavulanic acid and lowers the chance of nausea and stomach upset. Swallow the tablet whole with water.
Can I stop taking Augmentin 625 Duo when I feel better?
Take it for as long as your doctor prescribes, even if you feel better after a few days. Stopping early can let the infection come back and adds to antibiotic resistance. If side effects are troubling you, talk to your doctor.
Is Augmentin 625 Duo the same as amoxicillin?
No. It contains amoxicillin plus clavulanic acid. Clavulanic acid protects amoxicillin from enzymes made by some resistant bacteria, so the combination works against more types of bacteria than amoxicillin alone. It also causes diarrhoea more often. Your doctor decides which medicine an infection needs.
Why does Augmentin 625 Duo cause diarrhoea?
The antibiotic also kills some of the helpful bacteria in the gut. Mild diarrhoea is common, so drink plenty of fluids. Contact a doctor quickly if diarrhoea is severe or watery, has blood or mucus, or comes with cramps or fever, even weeks after the course.
Can I take Augmentin 625 Duo if I am allergic to penicillin?
No. It contains amoxicillin, a penicillin, and must not be taken by people who have had an allergic reaction to penicillins or cephalosporins. Tell every doctor and pharmacist about any antibiotic allergy.
Can I take Augmentin 625 Duo during pregnancy or breastfeeding?
Only if a doctor judges it necessary. Long experience has not shown a clear risk of birth defects, but one study linked it with a bowel problem in newborns when used after the waters broke early. Small amounts pass into breast milk, so watch the baby for diarrhoea, thrush or rash.
Can Augmentin 625 Duo affect the liver?
Rarely. It can cause hepatitis or jaundice, more often in older people, men and those on long courses, and signs can appear up to several weeks after the course ends. Get medical advice for yellow skin or eyes, dark urine, pale stools or tummy pain. People who had jaundice with it before must not take it again.
Can I drink alcohol while taking Augmentin 625 Duo?
No direct interaction between alcohol and amoxicillin with clavulanic acid is listed on the label. Alcohol can still add to stomach upset and make you feel worse while you are unwell, so ask your doctor.
Is Augmentin 625 Duo banned in India?
No. The combination of amoxicillin 500 mg and clavulanic acid 125 mg is approved and sold on prescription in India. Some other combinations that contain amoxicillin, such as amoxicillin with dicloxacillin and lactobacillus, have been banned.
